UDP-alpha-D-galactose + N-acetyl-beta-D-glucosaminylglycopeptide = UDP + beta-D-galactosyl-(1->4)-N-acetyl-beta-D-glucosaminylglycopeptide.
  • Terminal N-acetyl-beta-D-glucosaminyl residues in polysaccharides, glycoproteins and glycopeptides can act as acceptor.
  • High activity is shown toward such residues in branched-chain polysaccharides when these are linked by beta-1,6 links to galactose residues; lower activity toward residues linked to galactose by beta- 1,3 links.

UDP-alpha-D-galactose + D-glucose = UDP + lactose.
  • The enzyme is a complex of 2 proteins A and B.
  • In the absence of the B protein (alpha-lactalbumin) the enzyme catalyzes the transfer of galactose from UDP-galactose to N-acetylglucosamine (cf. EC 2.4.1.90).

    N-acetyl-beta-glucosaminyl-glycoprotein 4-beta-N- acetylgalactosaminyltransferase. [EC: 2.4.1.244]
    UDP-N-acetyl-alpha-D-galactosamine + N-acetyl-beta-D-glucosaminyl group = UDP + N-acetyl-beta-D-galactosaminyl-(1->4)-N-acetyl-beta-D-glucosaminyl group.
    • The enzyme from human can transfer N-acetyl-D-galactosamine (GalNAc) to N-glycan and O-glycan substrates that have N-acetyl-D-glucosamine (GlcNAc) but not D-glucuronic acid (GlcUA) at their non-reducing end.
    • The N-acetyl-beta-D-glucosaminyl group is normally on a core oligosaccharide although benzyl glycosides have been used in enzyme- characterization experiments.
    • Some glycohormones, e.g. lutropin and thyrotropin contain the N-glycan structure containing the N-acetyl-beta-D-galactosaminyl- (1->4)-N-acetyl-beta-D-glucosaminyl group.
