Functional Families

Overview of the Structural Clusters (SC) and Functional Families within this CATH Superfamily. Clusters with a representative structure are represented by a filled circle.

Superfamily: Metal-sensitive repressor, helix protomer

This superfamily includes metal-sensitive repressors, involved in resistance to metal ions. The overall protomer fold consists of three alpha-helices of varying lengths, organised in a closed tetrameric assembly. Members of this superfamily bind copper, nickel or cobalt ions via conserved cysteine and histidine residues. In the absence of metal ions, these proteins bind to promoter regions and repress transcription. When bound to metal ions they are unable to bind DNA, leading to transcriptional derepression PMID:17143269,PMID:20395270,PMID:17186148,PMID:17293508,PMID:16956381.
