The iron-sulfur flavoenzyme adenylylsulfate (adenosine 5'-phosphosulfate, APS) reductase catalyzes reversibly the reduction of APS to sulfite and AMP. It consists of two different subunits, alpha and beta, that are arranged as an alpha2beta2 heterotetramer. The alpha-subunit harbors the FAD cofactor and comprises an FAD-binding, a helical, and a capping domain; each of them an alpha/beta structure. This overall architecture classifies APS reductase as member of the fumarate reductase family.

The beta subunit consists of a bacterial ferredoxin-like segment carrying two [4Fe-4S] clusters, a three-stranded antiparallel beta-sheet, and a tail. Structural domains comprising this superfamily share the structure of the C-terminal region of this subunit which includes the beta-sheet and the C-terminal tail PMID:16503650,PMID:19820092,PMID:11842205.
