".

Functional Families

Overview of the Structural Clusters (SC) and Functional Families within this CATH Superfamily. Clusters with a representative structure are represented by a filled circle.

Superfamily: Filoviridae VP35, C-terminal inhibitory domain, beta-sheet subdomain

The filoviridae are a group of viruses that cause haemorrhagic fevers with a high mortality rate. The family currently contains three viruses: Ebola virus sp., Lake Victoria marburgvirus and Reston ebolavirus, named after their corresponding outbreak regions.

This entry includes the Ebola VP35 protein, which acts as a component of the viral RNA polymerase complex, a viral assembly factor, and an inhibitor of host interferon (IFN) production. This superfamily represents the C-terminal beta-sheet subdomain of the inhibitory domain located at the C-terminus of VP35.

VP35 contains an N-terminal coiled-coil domain required for its oligomerisation and a C-terminal dsRNA-binding region (inhibitory domain, IID). The C-terminus mediates the dsRNA binding activity, which is critical for viral suppression of innate immunity and for virulence PMID:19122151.

The VP35 IID structure is organised into 2 subdomains: an N-terminal alpha-helical subdomain and a C-terminal beta-sheet subdomain. The beta-sheet subdomain (residues 294-340) is composed of a 4-stranded mixed beta-sheet and a short helical segment PMID:19122151.
