Specificity close to that of papain. As compared to cathepsin B, cathepsin L exhibits higher activity toward protein substrates, but has little activity on Z-Arg-Arg-NHMec, and no peptidyl-dipeptidase activity.
  • A lysosomal enzyme that is readily inhibited by the diazomethane inhibitor Z-Phe-Phe-CHN(2) or the epoxide inhibitor E-64.
  • Belongs to peptidase family C1.

Cathepsin K. [EC: 3.4.22.38]
Broad proteolytic activity. With small-molecule substrates and inhibitors, the major determinant of specificity is P2, which is preferably Leu, Met > Phe, and not Arg.
  • Prominently expressed in mammalian osteoclasts, and believed to play a role in bone resorption.
  • Belongs to peptidase family C1.

Cathepsin S. [EC: 3.4.22.27]
Similar to cathepsin L, but with much less activity on Z-Phe-Arg-|-NHMec, and more activity on the Z-Val-Val-Arg-|-Xaa compound.
  • A lysosomal cysteine endopeptidase that is unusual among such enzymes for its stability to neutral pH.
  • Belongs to peptidase family C1.

Cathepsin V. [EC: 3.4.22.43]
The recombinant enzyme hydrolyzes proteins (serum albumin, collagen) and synthetic substrates (Z-Phe-Arg-NHMec > Z-Leu-Arg-NHMec > Z-Val- Arg-NHMec).
  • Maximally active at pH 5.7 and unstable at neutral pH.
  • Compound E-64, leupeptin and chicken cystatin are inhibitors.
  • Belongs to peptidase family C1.
