According to available literature members of this superfamily share structure of the enzyme 6-Pyruvoyl tetrahydrobiopterin synthase (PTPS), involved in the second of three steps in tetrahydrobiopterin synthesis. This domain is annotated in InterPro as a catalytic domain (INTERPRO:IPR007115).

It is important to note that the function of this domain was thought to be the same in the bacterial branch of the superfamily but, many are now taken to be QueD, and enzyme of queuosine biosynthesis. Queuosine is a hypermodified base in the wobble position of some tRNAs in most species (INTERPRO:IPR007115).
