Structural domains comprising this superfamily share the structure of DndE, a member of the highly conserved gene cluster dndA/iscS-dndBCDE that is widespread across bacterial and archaeal species involved in DNA phosphorothioation (PT). PT is a type of DNA backbone modification where a non-bridging oxygen atom of the phosphodiester bond is replaced with a sulfur atom in Rp (rectus for plane) configuration PMID:26539172. DndE is neither a sulfotransferase nor a NCAIR synthase analogue, but a possible nicked dsDNA binding protein, process which might be essential for DNA PT modification PMID:22525332.
