Superfamily: Tymovirus endopeptidases

Turnip yellow mosaic virus, or TYMV, is a member of the alphavirus-like supergroup of viruses and is a good model system for postitive-stranded RNA virus membrane bound replication. TYMV encodes a precursor replication polyprotein that is processed by the endoproteolytic activity of its internal cysteine proteinase domain. It also displays ubiquitin hydrolase (DUB) activity which is a key factor for the interaction of the virus with its host (enables counteracting the ubiquitin-proteasome system).

This entry represents the cysteine proteinase domain which exhibits a papain-like fold. The fold consists of two subdomains with the active site between them. One subdomain consists of a bundle of helices, with the catalytic Cys at the end of one of them, and the other subdomain is a beta-barrel with the active site His and Asn (or Asp).
