This superfamily contains mainly functionally uncharacterised proteins found in bacteria, typically between 99 to 124 amino acids in length.

The most representative domain was characterised in Rv0543c - a monomeric bundle of five alpha-helices rigidly held together by a largely conserved group of hydrophobic amino acid side chains. It irreversibly unfolds upon heating with Tm of 62.5C. The fold observed for Rv0543c may be unique for Rv0543c and the other members of the DUF3349 superfamily.

According to literature, the DUF3349 proteins are predicted to be good drug targets because they are predominately in the Mycobacterium and Rhodococcus bacterial species.
